| Study Design Background |
Cyclic nucleotides regulate adipogenesis and adaptive thermogenesis, with phosphodiesterases (PDEs) controlling their intracellular levels. PDE5A specifically modulates cGMP turnover in adipocytes. Although PDE5A inhibition has been explored in diabetes, its role in systemic metabolism remains unclear. This study used Pde5a knockout mice to investigate how PDE5A deficiency affects adipose tissue biology, energy homeostasis, and metabolic responses under normal and high-fat diet conditions.

| Study Description |
This study investigates the effects of PDE5A deficiency on adipose tissue biology and whole-body energy homeostasis using Pde5a knockout mouse models. Phenotypic, histological, and metabolic assessments were performed under chow and high-fat diet conditions, focusing on brown adipose tissue activation, white adipose tissue browning, hepatic lipid accumulation, and glucose metabolism. Mechanistic analyses explored cGMP and cAMP–PKA signaling pathways underlying metabolic reprogramming.
